Hydrophilic and hydrophobic interactions, which control the mobility of
molecules on the interface and their adsorption/desorption dynamics, are
important factors in varioys molecular processes and extensely studied
from the viewpoint of fundamental science as well as applications such
as the effective purification/separation of proteins and medicines. In
the present paper, single molecule tracking (SMT) using total internal
reflection fluorescence microscopy (TIRFM) was applied to the elucidation
of the translational diffusion and adsorption/desorption dynamics of amphiphilic
molecules with different length of alkyl chains on the surface of fused
silica modified with a trimethylsilyl group in water. The detailed statistical
analysis of trajectories of the amphiphilic molecules revealed that the
diffusion of the surface has two independent diffusion modes. Analysis
using Arrhenius plots of the temperature dependence of diffusion coefficients
provided the activation energy of the surface diffusion, indicating the
diffusion could be explained by a “hopping” model involving
alternate adsorption and desorption. In my presentation, I am going to
discuss the significance of the single molecule detection method by comparing
the present single molecule measurement method and conventional ensemble
measurement techniques as well.
界面における親・疎水性相互作用は、界面における分子の移動度や吸・脱着ダイナミクスを支配する重要な分子間相互作用であり、またその理解は、タンパク質・薬剤の高効率精製法の開発等にもつながるため応用上も重要な研究テーマである。今回紹介する論文では、蛍光色素(BODIPY)で修飾したアルカン酸分子の、表面をメチル化した溶融石英と水との固液界面における吸脱着及び表面拡散ダイナミクスを内部全反射顕微鏡(TIRFM)を用いて一分子ごとに追跡した研究結果が報告されている。
軌跡の詳細な統計的解析の結果、表面拡散は2成分の拡散係数を持つ異常拡散であることが明らかとなった。また拡散係数の温度依存性からこの表面拡散は活性化エネルギーを持つことが示され、アルカン酸分子は吸脱着を繰り返すホッピングにより表面を移動することが明らかとなった。講演では本論文で用いられた測定手法と従来の集団測定との比較も行い、単一分子レベルの測定で得られる情報の意義についても議論する。
