Fluorescent microscopy is one of the common methods to observe microstructures. Non-destructive internal observation provided by the optical imaging method has indeed contributed to the advancement in biological and chemical sciences by elucidating the mechanisms/dynamics of biological functions as well as micro-chemical systems. The conventional optical microscopy can however observe small structures at a diffraction-limited resolution that is about a half of wavelength, ca. 200 nm when visible light is used for imaging. Since the formulation of the diffraction limit by Abbe in the 19th century, there has been a barrier to observe nanometer-sized structures by optical microscopy. In recent years, however, several optical imaging methods, so-called super-resolution microscopy, with spatial resolutions beyond the diffraction limit have been developed and they are now employed to observe nanostructures. In my presentation, I am going to show previous approaches to attain super-resolution optical imaging, such as STED microscopy,[1] PALM,[2] and STORM[3] at the introductory part. Then I am going to show our original approach to achieve super-resolution imaging on the basis of the fluorescence photoswitching. The main part of my talk will start with the principle of the localization of excited states of photo-switchable fluorescence dyes beyond the diffraction limit, followed by the experimental data of the new imaging method with the discussion on its attainable spatial resolution and possible applications.
光学顕微鏡は最も一般的なマイクロ構造体観測法の一つである。サンプルの内部構造が非破壊・非接触で観察可能であり、生体組織の機能や微小領域における種々の化学プロセスのメカニズム・ダイナミクス解明に役立つ多くの情報を提供してきた。一方で、光学顕微鏡の空間分解能は光の回折限界のために半波長程度(可視光観察の場合で200 nm程度)に限定され、19世紀のアッベによる定式化以降、回折限界サイズより微小なナノ構造を光で観察することは非常に困難な課題であった。しかし近年、回折限界を超えた空間分解能を有する顕微鏡法(超解像顕微鏡法)が種々開発され、光によるナノ構造の観察が可能となっている。本発表では、まず研究の背景として代表的な超解像顕微鏡法、STED,[1] PALM,[2] STORM[3]について概説する。次いで我々が近年開発中の、蛍光スイッチング分子を用いた新たな超解像顕微鏡法についてその詳細を紹介する。蛍光のON/OFFスイッチングを利用して超解像蛍光イメージングを実現する原理について解説し、次に実際のイメージング結果を示すと共にその性能を評価する。得られたデータに関して、到達可能な分解能や応用可能性などに関して議論する。
